Alexei and his sisters

How was Alexei's relationship with OTMA? The majority of his sisters were much older than he.
Who was closer to him? Did he spent many time with they?

As far as I know, he was closest with Anastasia (naturally, since she was only about three years older than him)...

I read something that in the time of the exile he spent a lot of time with Olga. Is it true?

During captivity, he got closer with Olga, while Tatiana used to help him moving to bed and spent most of time with him.

I suppose yes. His sisters worshipped him in any way, it is fact, especially during captivity -- they played with sledge rides during winter of 1917 altogether, as a view of playing.

He almost certainly did -- what siblings don't? -- but I can't recall reading about any specific incidents.

That's possible. Also, Aleksei's daily routine was somewhat separate from OTMA's -- his bedroom did not adjoin theirs, and he had his own classroom. It wasn't uncommon for him to take meals separately from his sisters. During the war the girls were often visiting/working in their hospitals, and Aleksei visited Stavka for weeks at a time with his father. So during their teen and pre-teen years there wasn't as much opportunity for contact between the imperial siblings, which could mean there was also less friction than most teenaged siblings experience.

I've never found any complaints in Aleksei's letters or diaries. In fact, he had more access to friends his own age than OTMA did.
